Could those spots also be on the lens?

Details look great but so little color. Purposely?

maybe i tried to clear too much from a very dark image

At the f/16 you were shooting, yes. Their circular shape suggests that they are actually on the lens somewhere.

In don’t think so. I get such spots when my sensor is dirty. But who knows.

A good way to spot sensor dust is to stop down the lens as much as possible, and in manual focus mode (you want a blurry photo) take a photo of a uniformly illuminated wall. I often use a long exposure time (we are stopped down, anyway), and move and twist the camera to even out the exposure. Then, in the raw processor, add tons of contrast.
